Like his father Johann Sebastian Henne, who had held the pastor's position in Gunsleben and Hammersleben in the Principality of Halberstadt since 1706, Samuel David Ludwig Henne was also a pastor. In the mid-1750s, Henne started a fruit tree nursery. This consisted of 15,000 trees and he grafted 2,000 trees every year. He wrote down the experiences he gained there in his book Instructions on how to properly maintain a nursery of fruit trees , which was first published in 1769 and which appeared in further editions in 1772, 1773, 1774 and 1791. Henne published the first editions of his work anonymously. At the end, Henne only added the Latin motto Soli Deu Laus Honorque (in German Praise and Honor be to God ), the first letters of which correspond to his initials.

Henne also bred a high-yielding hazelnut variety and the Gunsleber thick apple . His son was the engraver Eberhard Siegfried Henne .
