Stanley T. Crooke

Stanley T. Crooke (born March 28, 1945 ) is a pharmacologist and entrepreneur ( Ionis Pharmaceuticals in Carlsbad , California ).

Crooke is best known for developing antisense oligonucleotides . The preparations developed by Ionis from this class of active ingredients with (partly only temporary) approval include Mipomersen (trade name Kynamro ) for the treatment of homozygous familial hypercholesterolemia , Fomivirsen (Vitravene) for the treatment of infections with the cytomegalovirus (CMV) in immunodeficiency , Nusinersen (Spinraza ) for the treatment of spinal muscular atrophy , Inotersen (Tegsedi) for the treatment of polyneuropathy in hATTR and Volanesorsen (Waylivra) for the treatment of familial chylomicronemia (FCS).

Crooke comes from Indianapolis and grew up in difficult family circumstances. He graduated from Baylor College of Medicine , where he received a Ph.D. from Harris Busch. and earned an MD while also working as an intern. After working at Bristol Laboratories (now Bristol-Myers Squibb ), where he helped set up the oncology division, and as head of research and development at SmithKline (now GlaxoSmithKline ), Crooke founded the pharmaceutical company ISIS together with C. Frank Bennett in 1989 (later renamed to Ionis) to develop antisense oligonucleotides there, initially without much success. It was not until 2008 that the first preparation, mipomersen, received approval from the Food and Drug Administration (FDA).

Crooke was - together with Ryszard Kole - the Massry Prize 2019 awarded. According to the Scopus database , Crooke (as of November 2019) has an h-index of 73.

Stanley Crooke is married to Rosanne Crooke, who is also a pharmacologist with Ionis.
