After initially working in the diamond industry, Querido opened his own bookstore on the Binnenamstel in Amsterdam in 1898 and also began publishing books. In 1911 he moved the business to Bloemendaal as a mail order bookshop , but had to close it in 1913 for economic reasons. In 1915 he founded the publishing house Em. Querido's Uitgeversmaatschappij NV, based in Keizersgracht 333 in Amsterdam, and became a successful entrepreneur. In 1934 he founded the Salamander series, the first paperback series in the Netherlands.

Querido and his wife Jane Querido-Kozijn retired to Laren , where they had been living privately since 1929. In 1943 they went to a hiding place in neighboring Blaricum , but were betrayed, taken to Sobibor and murdered there.
