Plütschau attended the Friedrichswerder Gymnasium in Berlin under its rector Joachim Lange and then studied theology at the University of Halle . There he came into contact with August Hermann Francke , the founder of the Glauchaschen orphanage . When the Danish King Friedrich IV asked about potential missionaries, Plütschau and his fellow student Bartholomäus Ziegenbalg were suggested by Joachim Lange. When it was sent to the Danish colony of Trankebar on the southeast coast of India on November 29, 1705 , the Danish-Hallesche Mission came into being .

The missionaries had already started to learn Portuguese during the crossing , the local lingua franca . In April 1706 they spent a few days at the Cape of Good Hope in order to be able to relax. After arriving in southern India on July 9, 1706, they also immersed themselves in the Tamil language ("Malabar language"). As the work increased, the missionaries shared their ward duties. Plütschau concentrated on the parishioners who spoke Portuguese, and Ziegenbalg, because of his special language skills, on the Tamil-speaking community. However, Plütschau had to return to Germany as early as 1711 for health reasons. There he published the small catechism in Tamil and taught mission candidates in this language. In 1714 he received the pastor's post as a Lutheran pastor in Beidenfleth, which was then ruled by the Danish king, where he worked for 36 years until 1750.
