ABCG1 is one of two proteins in the cell membrane of mammals that pumps cholesterol out of the cell using ATP , where it floats on the membrane and is taken up by HDL particles ( reverse cholesterol transport ). It belongs to the ABC transporter family . Oxidation products of cholesterol accumulate in mouse endothelial cells without ABCG1. ABCG1 also plays the main role in cholesterol transport in macrophages and monocytes . In other cells this is taken over by ABCA 1 .

In type 2 diabetes mellitus patients, the expression of ABCG1 in monocytes is reduced, which contributes to atherosclerosis , similar to the activity of the enzyme 12/15 lipoxygenase . However, overexpression leads  to apoptosis via translocation of phosphatidylserine and caspase 3 activation .
