At the Summer Olympics in Sydney that same year , however, he lost his first fight against Vladimir Sidorenko on points.

He has been boxing in the professional camp since March 2001 and is under contract with Golden Boy Promotions . He won his first 24 fights in a row, 23 of which by knockout. The defeated opponents also included the World Cup challengers Jesús Salvador Pérez (23-3), César Figueroa (26-3), Emmanuel Lucero (22-2) and Julio Gamboa (28-8).

In his 25th professional fight on February 17, 2005, he lost on points to five-time world champion Celestino Caballero (20-2), but won his next fight by knockout against Ricardo Barajas (23-4). On October 29, 2005 he finally boxed in Tucson for the WBO World Cup against Sod Kokietym (25-0) and won unanimously on points. For the first time he defended his title in May 2006 by knockout in the second round against Gerson Guerrero (30-6). He also won the rematch against Sod Kokietym with a knockout in the first round.

In October 2006 he prevailed by tKo in the eighth round against Al Seeger (27-1) by winning unanimously on points against Gerry Peñalosa (51-5) in March 2007 . In August of the same year he won by tKo in the first round against Rey Bautista (23-0) and in September in a non-title fight by knockout in the fifth round against Reynaldo López (28-4). In December he defended his title for the sixth and last time against Eduardo Escobedo (21-2) by unanimously scoring.

On June 7, 2008 he lost his WBO title by a surprising tKo defeat in the first round to the Puerto Rican Juan Manuel López (21-0), after Ponce de León had to accept two heavy knocks and the referee from the fight was taken. He then switched to featherweight and won his next seven fights, including prematurely against Orlando Cruz (16-1), Antonio Escalante (24-2) and Sergio Manuel Medina (35-3).

On March 5, 2011 he lost controversially on points against Adrien Broner (19-0), as well as on September 10, 2011 by technical decision against Yuriorkis Gamboa (20-0); Due to an injury to Ponce de León's forehead from an unintentional headbutt by Gamboa in the infight, the fight was stopped in the eighth round and the scores on which the Cuban was in the lead were evaluated.

With two subsequent wins against Omar Estrella (15-3) and Eduardo Lazcano (24-2) he got a WBC World Cup chance against Jhonny González (52-7) and won the title early in the eighth round.

On May 4, 2013, he lost his world title through a tKo defeat in the ninth round to Abner Mares (25-0). On March 15, 2014 he also lost again to Juan Manuel López (33-3) by tKo in the second round.
