Daniele Barbaro's most important work is probably the translation of the books by the ancient architect Vitruvius published in 1556 . He had been advised by Andrea Palladio , whose illustrations he used and who then elevated Vitruvius - in his own interpretation - to the general architectural standard. Palladio also advised Daniele Barbaro and his brother Marcantonio Barbaro on the construction of their Villa Barbaro in Maser .

Barbaro also wrote numerous writings on art, mathematics, science, and optics. The use of a ground lens in a camera obscura is described for the first time in the work “ La pratica della persepttiva ” by Daniele Barbaro from 1569. Leonardo da Vinci had already recorded this knowledge in one of his sketchbooks , but these were not published during his lifetime.
