Elisabeth was the eldest daughter of Landgrave Moritz von Hessen-Kassel (1572–1632) from his marriage to Agnes (1578–1602), daughter of Count Johann Georg zu Solms-Laubach . Her godmother was Queen Elizabeth I of England , who was represented on the occasion by the Earl of Lincoln . The baptism of the princess was one of the most elaborate celebrations at the court in Kassel, on the occasion of which one of the last knight games in Europe was held. The engraver and historiographer Wilhelm Dilich summarized the event between 1598 and 1601 in a documentation. The volume is now in the city museum of Kassel.

Initially intended to be the wife of the Swedish Duke Karl Filip , this marriage project fell apart. She was then engaged to Friedrich Heinrich von Orange , but the latter broke the engagement after he had failed with his considerable dowry demands.

Elisabeth married Duke Johann Albrecht II of Mecklenburg on March 25, 1618 in Kassel , who already had four children from his first marriage. The highly musical duchess created the Güstrow court orchestra and continued to write.

Elisabeth had pledged no children from her marriage, her marriage property, to the office and city of Dargun , and after her death fell back to Hesse.
