He was the son of Count Franz Anton von Hohenzollern-Haigerloch (1657–1702) and Maria Anna Eusebia von Königsegg-Aulendorf . He himself remained unmarried. One of his brothers was Franz Christoph Anton von Hohenzollern-Sigmaringen .

In 1706 he entered the Cologne Cathedral Chapter . He was canon in Speyer from 1714 to 1726 . In 1725 he was also canon in Strasbourg . In Cologne he was choir bishop between 1724 and 1727 and became vice dean in 1727. In 1731 he became cathedral dean . In 1733, Elector Clemens-August made him the successor of the previous first minister, Ferdinand von Plettenberg . He also took over the office of chief steward. However, his political influence was significantly less than that of his predecessor.
