During his time as Lord Proprietor, however, between 1763 and 1767 he surveyed the Mason-Dixon Line , the border between Pennsylvania and Maryland that is still valid today , in order to settle disputes with the Penn family from Pennsylvania.

On March 9, 1753 he married Lady Diana Egerton (1732-1758). In 1768 he was charged with rape in England but acquitted. He then left England and traveled through Europe until his death. Frederick Calvert died without any legitimate descendants, the title was no longer valid.

His illegitimate son Henry Harford (1758-1834) from his relationship with Hester Whelan was his successor as Lord Proprietor of the colony of Maryland.
