Bozzuffi began his career in 1955 with supporting roles in films such as Le Fils de Caroline chérie by Jean Devaivre and Gas-Oil (1955), then starred in The Second Breath (1966) by Jean-Pierre Melville , in Costa-Gavras ' political thriller Z (1969 ) and in the thriller Focal Point Brooklyn from 1971. During this time he also worked on several comedies by Claude Lelouch , La Vie, l'Amour, la Mort (1969) and Le Voyou (1970).
As a film star, he had acquired the image of the cold-blooded killer, for example in Lucio Fulci's thriller The Syndicate of Horror (1980) in the role of "Marseiller".

Bozzuffi played the role of Gaetano Bonanno, clan chief of the criminal organization Camorra in Naples in the 4-part Italian television film Herz aus Stein by Lucio de Caro and Steno (Stefano Vanzina), which was shown on ARD in July 1988.
