He made important discoveries about the expression and regulation of perforin , was able to clone novel granzymes and outline the killing effect of granules on cancer cells and to elucidate the natural anti-tumor effect of Tumor Necrosis Factor Related Apoptosis Inducing Ligand (TRAIL). He also showed that the course of cancer can be shaped by the selection of cancer cells that evade the immune system (immunoediting) , paving the way for the development of immune checkpoint inhibitors . Other important contributions deal with the function of NK cells and NKT cells or CD73 and CD96 as target molecules of cancer immunotherapy .
Smyth is the most cited immunologist in Australia. It has (as of September 2018) an h-index of 140.
