Mark Ptashne
Mark Steven Ptashne (born June 5, 1940 in Chicago ) is an American molecular biologist and professor at the Memorial Sloan-Kettering Cancer Center .
Life
Ptashne earned a bachelor's degree in chemistry from Reed College in Portland , Oregon in 1961 and a Ph.D. in molecular biology from Harvard University in Cambridge , Massachusetts . In 1971 he received a professorship there in molecular biology and in 1997 a professorship at the Memorial Sloan-Kettering Cancer Center , New York City .
Ptashne is also a passionate violinist , owner of a Stradivarius violin and has made CD recordings with this instrument.
Act
Ptashne carried out fundamental work on the isolation of repressors , which play an important role in the regulation of cellular processes. He succeeded in elucidating the interaction between repressors and other proteins (e.g. Cro ), which in turn interact with the DNA . He was able to describe the three-dimensional structure of the repressor and interacting DNA. While first work was carried out on the bacteriophage Lambda and described the regulation between the lysogenic cycle and the lytic cycle , Ptashne was later able to describe similar processes in yeasts and higher organisms. Due to his work, the yeast protein GAL4 is one of the best-studied transcription factors . The discovery that transcription factors usually consist of separable domains that mediate DNA binding or the interaction with coactivators and corepressors was the prerequisite for many important molecular biological techniques, such as the yeast two-hybrid system .
