The brothers Joe and Paul Fabrini are truck drivers. His own truck has not yet been paid off and Joe dreams of founding his own haulage company. He works for this dream, but his brother Paul would rather give up the vagabond life and settle down with his wife Pearl. On the night when the last loan installment for the truck is to be earned, Paul falls asleep behind the wheel and speeds the truck over a cliff. Paul survives but loses an arm. The truck is completely destroyed and Joe's dreams are gone. Joe then accepts the offer from Ed Carlsen, a freight forwarder, and starts working in his office. Lana Carlsen, Ed's wife, is in love with Joe Fabrini, but realizes that Joe is in love with Cassie Hartley. When Joe lets her down , Lana murders her drunken husband in frustration. Initially, the murder is viewed as an accident.

After the death of her husband, Lana offers Joe a stake in the company. However, when she realizes that Joe actually wants to marry Cassie Hartley, she accuses Joe of instigating the murder of her husband. The police initially believe her and accuse Joe. Plagued by a trauma caused by the murder of her husband, Lana finally loses her mind in court and a psychiatrist is called in. Joe is released and returns to Cassie and the company.

Night En route was based on the novel The Long Haul by AI Bezzerides . The film was shown in Germany not in cinemas, but was first broadcast on 22 July 1967 at the television.
