Protein E6 is an oncogenic protein of the human papillomavirus 16.

Protein E6 brings resting cells to cell division by overcoming the checkpoints of the cell from the G 0 phase to the S phase . In addition, it inhibits apoptosis and inhibits the immune response . Combined with E7 transformed it host cells .

Protein E6 binds to the ubiquitin ligase UBE3A (synonym E6-AP) and inactivates p53 and p73 by degradation in the 26S proteasome . This increases DNA damage and chromosomal aberrations , which ultimately result in the development of cancer . The protein complex of E6 and UBE3A binds various other cellular proteins and leads them to degradation in the proteasome, for example DLG1 , NFX1 , BAK1 , FADD- containing proteins, procaspase 8 , IRF3 and TYK2 . NFX1 is an inhibitor of the gene expression of the hTERT , which is why the degradation of NFX1 increases the formation of hTERT, which leads to an immortalization of the cell. BAK1, FADD proteins and the procaspase 8 initiate apoptosis, which is why their breakdown leads to an inhibition of apoptosis. IRF3 and TYK2 are involved in the immune response, which is why their breakdown leads to a reduced immune response due to the inhibited induction of genes of the immune reaction or the activation of Jak-Stat , which inhibits the formation and effect of interferon . The C terminus of protein E6 binds to the PDZ domain of PDZ-containing proteins.
