Rho kinase 1 (gene: ROCK1 ) is an enzyme that activates several other enzymes through phosphorylation as soon as it has  bonded with the small G protein Rho A. It is therefore an essential part of signal transduction . Rho kinase is involved in regulating various cell functions such as the contraction of smooth muscle , the organization of the actin - cytoskeleton , the cell adhesion , the cell migration , of cytokinesis , the cell proliferation and the migration of inflammatory cells.

The Rho kinases evolved with the bilateral animals . In humans, Rho kinase 1 is located in the cytoplasm , in the membrane of the Golgi apparatus and in the centrioles during the S phase , but especially in platelets . Freshly formed Rho kinase 1 inhibits itself by binding the catalytic center to the C terminus. This inhibition is lifted by binding to Rho (reversible) or by severing the end (irreversible). This separation is catalyzed by the enzyme caspase-3 , especially during cell death , which ultimately causes the membrane to bulge ( blebbing ).

Fasudil hydrochloride is a specific inhibitor of Rho kinase.
