Charles O'Neill attended Dickinson College in Carlisle until 1840 . After a subsequent law degree and his admission as a lawyer in 1843, he began to work in Philadelphia in this profession. At the same time he embarked on a political career. He later became a member of the Republican Party founded in 1854 .

In the congressional elections of 1862 O'Neill was the second electoral district of Pennsylvania in the US House of Representatives in Washington, DC chosen, where he succeeded the on March 4, 1863 Democrats Charles John Biddle took. After three re-elections, he was able to complete four legislative terms in Congress by March 3, 1871 . These were shaped by the events of the civil war and its consequences. Since 1865, the work of Congress has been overshadowed by tension between Republicans and President Andrew Johnson , which culminated in a narrowly unsuccessful impeachment trial.

In 1870, O'Neill was not re-confirmed. In the elections of 1872 he was then re-elected to Congress in the second district of his state, where he replaced John Vaudain Creely on March 4, 1873 , who had been his successor two years earlier. After ten re-elections, he could remain in Congress until his death on September 25, 1893. From 1883 he represented the third district of his state there as the successor to Samuel J. Randall .
