Masson trichrome stain

from Wikipedia, the free encyclopedia
Masson trichrome staining of rat lung tissue, with connective tissue (blue), cell nuclei (red / purple) and cytoplasm (red / pink)

The Masson's trichrome staining , histological trichrome staining method .

properties

Most variants of Masson's trichrome stain produce red-colored keratin and muscle fibers , blue or green collagen and bones , pink cytoplasm and brown-black cell nuclei .

After fixation, the objects to be stained are immersed in Weigert's iron and hematoxylin solutions to stain the cell nuclei . The first Weigert solution contains ferric chloride in dilute hydrochloric acid , the second consists of hematoxylin in 95% ethanol and the third of potassium hexacyanoferrate in alkaline sodium borate solution. This is followed by cytoplasmic staining with acid fuchsin and Ponceau 2R in an aqueous acetic acid solution. In the variant of Masson staining according to Lillie, Biebrich scarlet is used as an alternative acidic dye . The next solution contains phosphomolybdic acid . Finally, the protein fibers are colored with light green SF or Fast Green FCF . Alternatively, a blue coloration with methylene blue , water blue or aniline blue can be used.

variants

The Lillie trichrome stain uses Biebrich's scarlet fever as the red dye . The Masson Trichrom & Verhoeff stain is a staining combination that is used to stain blood vessels and to differentiate between arteries and veins , as the Verhoeff stain stains elastin black. In the Masson-Goldner trichrome staining , hematoxylin is replaced with iron trioxyhematein according to Hansen, the other color solutions are diluted, phosphomolybdic acid is replaced by phosphotungstic acid and orange G is used to stain the erythrocytes .

history

Masson trichrome staining was developed in 1929 by Claude L. Pierre Masson (1880-1959). It is a further development of the Mallory trichrome stain .
