α-5'-phosphoribosyl-1'-pyrophosphate (PRPP) is a chemical compound . It is a ribose (a sugar) which is substituted on carbon atom 5 (C5) with a phosphate group and on C1 with a pyrophosphate group . Phosphoribosyl pyrophosphate occurs primarily as an intermediate in metabolism , for example in the biosynthesis of purine and pyrimidine nucleotides .
It is also required as a cosubstrate in the biosynthesis of NAD + / NADP + from nicotinic acid .
Phosphoribosyl pyrophosphate reacts with nicotinate, the anion of nicotinic acid, to form nicotinate mononucleotide with cleavage of pyrophosphate (PP i ).
PRPP is synthesized in the body from ribose-5-phosphate and ATP or deoxy-ATP. The reaction is catalyzed by the enzyme ribose phosphate pyrophosphokinase .
