Protein-RNA interaction

A protein-RNA interaction refers to the binding of a protein to RNA . This controls various processes of gene expression in cells .

Proteins that interact with RNA are called RNA-binding proteins, e.g. B. ribonucleoproteins , proteins of transcription and translation , RNA interference , miRNA -binding proteins, RNA-modifying enzymes such as RNases and the DNase Cas9 .

The interactions can be divided into sequence-specific and sequence-independent protein-RNA interactions. In the case of the sequence-specific binding proteins, the binding takes place at least partially to the ribonucleic bases ( adenine , cytidine , guanine , uracil ), while an interaction with the ribose - phosphate repeat units of the RNA backbone is sufficient for a sequence-independent binding .

Interactions defined in the same way are e.g. B. Protein-lipid interactions , protein-protein interactions, and protein-DNA interactions .

Verification procedure

Various methods can be used for characterization ( RNA sequence , RNA-binding proteins). Protein-RNA interactions can e.g. B. can be detected by immunoprecipitation , EMSA , RNA-Seq , RIP-Chip , ICLIP , CLIP-Seq or by PAR-CLIP . In addition, structure elucidation can be carried out by X-ray structure analysis or by NMR spectroscopy . Furthermore, the affinity of a binding can be determined by a ligand binding test, e.g. B. by filter binding test .
