She was the third daughter of Duke Theodor Eustach von Pfalz-Sulzbach and his wife Maria Eleonora Amalie, Countess of Hesse-Rheinfels-Rothenburg. The dukes of Pfalz-Sulzbach were a Palatine branch line of the Wittelsbach family .

Ernestine was brought up together with her older sister Franziska Christine von Pfalz-Sulzbach in the Essen monastery . Charles VI arranged the wedding with 48-year-old Wilhelm the Younger from Hessen-Wanfried. The marriage took place on September 19, 1719. The marriage remained childless. After the death of her husband in 1731, the widow initially lived at Rheinfels Castle, later she joined the Carmel of Neuburg an der Donau and finally became prioress of the monastery, where she received the religious name Augusta. According to Torsy-Kracht, "she was a role model for her fellow sisters in obedience, humility and love for poverty" . She died as the prioress of the convent in the name of holiness. Their bones were transferred to the Neuburg Court Church .

The older sister Maria Anna Amalia Auguste von Pfalz-Sulzbach (1693–1762) had lived as a Discalced Carmelite since 1714 in the Cologne monastery of St. Maria in the Kupfergasse . She also died in a reputation for holiness.
