Born as the son of Balthasar Glaß, who was initially active as a Schwarzburg rent clerk in Grüningen and Sondershausen and later became an official in Gehren, and his wife Anna Maria Müller, Glassius first began studying at the University of Jena . During his studies he switched to the University of Wittenberg and returned to the Jena Academy, where he became Johann Gerhard's favorite student and was accepted into his household.

Under his support, Glassius acquired the academic degree of a master’s degree in 1617 , was accepted as an adjunct at the philosophical faculty of the academy and in 1621 became professor of the Hebrew and Greek languages. In 1625 he was appointed superintendent in Sondershausen , received his doctorate in theology in Jena in 1625 and succeeded Gerhard in 1638 as professor of theology at the University of Jena.

In 1640 he accepted the offer of Duke Ernst I of Saxe-Gotha to take over the position of general superintendent of Saxe-Gotha, the office of court preacher, consistorial assessor and Ephorus at the Gotha grammar school. In this office he played a major role in the rebuilding of the church and school system in the duchy, which had fallen through the Thirty Years' War .

After he had headed the Jena Academy as Rector of the Alma Mater in 1625 and 1639 , he took over the visitation in his new office, participated in the development of biblical hermenautics and wrote catechetical, exegetical-homiletic and argendarian literature. After Gerhard's death he took part in editing the Weimar Bible. Due to his theological practice, which was based on the “pure teaching” of the Bible, he is considered a spiritual father of August Hermann Francke .

Glassius found his final resting place on Gotha Cemetery I ( Alter Gottesacker ) between Bürgeraue and Eisenacher Strasse. After the cemetery was closed in 1874, his grave slab was moved to the cloister of the Augustinian Church , where it can still be seen today.
