4-thiouridine is a chemical compound from the group of uridines . It is a rare nucleoside that contains the sulfur-containing base 4-thiouracil .
Occurrence
The compound is part of several transfer RNAs . It was discovered in Escherichia coli .
properties
4-thiouridine is a crystalline yellow solid that is soluble in water and methanol. It has a monoclinic crystal structure with space group C 2 (space group no. 5) .
use
4-Thiouridine is a thionucleobase used as an antisense agent and for RNA analysis, including RNA-RNA cross-coupling and RNA labeling. It is a photoreactive (cross-linking) uridine analogue that can be incorporated into RNA structures during phosphorylation to 4-thioUTP. The compound can be used to determine the specificity and kinetics of uridine cytidine - kinases to investigate, and is typically used to modify oligos, which are provided for RNA or RNA-protein structural studies. A 4-thio-rU modified RNA pentamer was used to study the effect of this modification on the codon-anticodon interaction when it is in the wobble position of the tRNA.
