Osten-Sacken received his first training from private tutors, and later he attended the Armand Richelieu Lyceum in Odessa .

He began his professional career in 1852 as an official in the Russian Foreign Ministry. During the Crimean War (1853-1856) he served at times as an official for special tasks of the commander-in-chief of the Russian army. His father did a great job defending Sevastopol during the war and was given the title of count for it in 1855. Since this also extended to his descendants, his son Nikolai was raised to the rank of count.

Nikolai von der Osten-Sacken was sent abroad for the first time in 1856. Initially, he was a member of the Russian embassies in The Hague , and later he worked in Madrid , Bern and Turin . Shortly before the beginning of the Franco-Prussian War of 1870/71, he came to Darmstadt as an envoy , where he stayed for eleven years. This was followed by a transfer as envoy to Munich and finally, from 1895, the climax and conclusion of his career, the post of Russian ambassador for the German Reich in Berlin, where he succeeded Count Paul Andreevich Shuvalov for almost seventeen years, until 1912, worked. In 1910 he was awarded the Order of St. Andrew , the highest order in the Russian Empire. In his capacity as the honorary chairman of the non-profit brotherhood of the holy Prince Vladimir , in connection with the office of ambassador , Osten-Sacken supported the building of Russian Orthodox churches in Germany.

Osten-Sacken 's wife was the princess Maria Ilyinichna Dolgorukova (1822-1908), who was married to the Russian ambassador to Spain, Prince MA Golitsyn. She was considered a patron of the arts, especially music. The Countess, who had once studied with Frédéric Chopin , was a patroness of the composer Seregei Bortkiewicz , the relative of one of her husband's employees.
