Specklin first learned the trade of silk embroidery, a prosperous craft that not only included embroidery with gold and silver threads, but also required designs and drawings for the same. He finished his apprenticeship in 1552 at the age of sixteen, followed by the usual years of traveling. In 1554 he was in Hungary , where he got to know the fortress building in Komorn and apparently began to work in this profession. In 1555 he was present at the construction of the new fortifications in Vienna . He made rapid progress in his new profession, became a site manager in Vienna and had his own construction sites to look after. In 1560 he was in Antwerp , then again in Vienna. In 1564 he returned to Strasbourg. There he worked in his old trade as a silk embroiderer, but also acquired knowledge of cartography and tried his hand at a map of the city of Strasbourg . Then he was traveling again, u. a. to Düsseldorf, Regensburg and Hungary. In 1573 he received an order from the Austrian Archduke Ferdinand to map the whole of Alsace ; he worked on it until 1576. The large map, printed on three plates, then appeared in 1577 (engraved by Franz Hogenberg). In 1576 he set out for Ingolstadt on behalf of the Bavarian Duke Albrecht V in order to renew the fortifications there; In 1577 he stayed in Ulm for similar purposes. In the same year, on October 5, 1577, he was appointed city architect of his hometown - for an annual salary of 250 guilders.

In addition to the design and implementation of a large number of construction works - also in other Alsatian cities, e.g. B. Ensisheim - he also wrote theoretical treatises (main work: Architectura von Vestungen , 1589) and wrote a Strasbourg chronicle based on the famous Schedelsche Weltchronik . At the age of 53 he died on October 18, 1589 in Strasbourg in the house of his niece Katharina, who was married to the printer Lazarus Zetzner .

Specklin was married to Susanna Wegranft (widowed entringer) since 1565. There were no children from the marriage. His legacy was shared between his brothers Josias, Zacharias and Jeremias and Specklin's niece Katharina.

The family name "Specklin" has been recorded in Strasbourg since 1399; Specklin called itself "Speckle", was sometimes also called "Speckel".
