D'Isoard attended the Small Seminary in Aix, where he met Joseph Fesch , Napoleon Bonaparte's uncle , but did not complete his theological training. During the reign of terror , he fled to Italy in 1794 to the court of the future King Louis XVIII. After the coup d'état of the 18th Fructidor V , he was again forced to flee to Italy. After Napoleon had established the consulate , he returned to accompany Joseph Fesch, who had been appointed French ambassador to the Pope, to Rome shortly afterwards. At his instigation, d'Isoard was appointed auditor of the Roman Rota in 1803 .

After Napoleon annexed the Papal States in 1809 and sent Pope Pius VII into exile in France, d'Isoard followed him, declining various offices offered to him by Napoleon, including senator. After the liberation of the Pope towards the end of Napoleon's rule, he returned to Rome and was briefly the French chargé d'affaires at the Holy See during the rule of the Hundred Days .

The new King Louis XVIII. ran his replacement as auditor of the Rota by Louis-Siffren-Joseph de Salamon without success .

D'Isoard, who had been dean of the Roman Rota since 1824, was ordained a priest in 1825 at the age of 58 . In the consistory of June 25, 1827 he was by Pope Leo XII. elevated to cardinal with the titular church of San Pietro in Vincoli . On July 6, 1828, he was appointed Archbishop of Auch ; the episcopal ordination took place on January 11, 1829 in Paris by Cardinal Jean-Baptiste de Latil . Charles X appointed the avowed monarchist Peer of France in 1829 . During his time as Archbishop of Auch, he took part in the papal elections of 1829 (election of Pius VIII. ) And 1830 to 1831 (election of Gregory XVI. ), In the latter conclave he was entrusted with exercising exclusive rights for France if necessary .

On January 15, 1833 Cardinal d'Isoard changed the titular church and became cardinal priest of Santissima Trinità al Monte Pincio . After rejecting the bishopric of Bordeaux once and that of Aix-en-Provence twice, in 1839 he accepted the appointment of Archbishop of Lyon to succeed Cardinal Fesch. Before he could take office, he died on October 7th of the same year in Paris of an inflammation of the chest.
