After a victory over Stephan Bonnar at Jungle Fight 1 on September 13, 2003 in Manaus , he inflicted his first defeat on the later UFC Middleweight Champion Rich Franklin at the Inoki Bom-Ba-Ye Festival 2003 . Machida then competed three times at K-1 , defeating Michael McDonald, Sam Greco and BJ Penn on March 26, 2005 in Saitama . After winning the Jungle Fight and against Vernon White in the World Fighting Alliance , Lyoto Machida made his successful debut in the Ultimate Fighting Championship at UFC 67 on February 3, 2007 . After three more victories in the UFC , including against Kazuhiro Nakamura , he fought on May 24, 2008 at UFC 84 victorious against Tito Ortiz . In the next fight, against Thiago Silva at UFC 94 on January 31, 2009, his victory was declared Knockout of the Night . He then went on May 23, 2009 at UFC 98 for the title fight for the Light Heavyweight Title against Rashad Evans . He defeated Evans and was able to defend the title on October 24 at UFC 104 against Mauricio Rua , but in the rematch on May 8, 2010 at UFC 113, Machida Rua was knocked out and had to deal with his first MMA defeat.

In his next fight, at UFC 123 on November 21, 2010, he lost to Quinton Jackson just on points. On April 30, 2011, Machida defeated Randy Couture at UFC 129 in the 2nd round by a spectacular knockout, whereupon Couture announced his sporting retreat. In December 2011, Machida was defeated by Jon Jones with a guillotine choke in the 2nd round. In 2013 Machida switched to middleweight division, where he took up the title fight against Chris Weidman after victories against Mark Muñoz and Gegard Mousasi on July 5, 2014 at UFC 175 , but lost it by unanimous decision. A win against CB Dollaway was followed by premature defeats against Luke Rockhold and Yoel Romero .

In April 2016, Machida tested positive for a steroid during a doping control and was banned for 18 months. After his suspension, Machida fought three times at the UFC, winning two fights against Eryk Anders and Vitor Belfort and losing one to Derek Brunson.
