The South Fork Township is about 60 km north of the Missouri River and about 85 km west of the Mississippi , which forms the border with Illinois .
The South Fork Township is located at 39 ° 10 ' N , 91 ° 52' W and extends over 120.15 km², which is spread over 118 km² of land and 2.15 km² of water.
The South Fork Township is located in the southern center of Audrain County and borders on Callaway County to the south . Within Audrain County, the South Fork Township is bordered by the Salt River Township to the west, the Prairie Township to the north and the Linn and Loutre Township to the east .

US Highway 54 runs through the South Fork Township as the eastern artery of the city of Mexico . All other roads within the township are subordinate and partly unpaved roads as well as inner-city connecting roads.
In South Fork Township, a Norfolk Southern Railway line , which runs from St. Louis to Kansas City , meets a Kansas City Southern line , which also runs from Springfield , Illinois to Kansas City.
