After her mother remarried, the family moved to Texas. She dropped out of high school after 9th grade because she said she "preferred to entertain classmates rather than an academic education." Notaro moved to Denver, where she briefly worked in the music industry, before moving to Los Angeles and pursuing her career as a stand-up comedienne.

From the mid-2000s she also appeared in film and television productions. She became known, among other things, for her role as a lesbian police officer Officer Tig in several episodes of the Sarah Silverman program .

In 2012, Notaro was diagnosed with breast cancer. She addressed her illness directly at a performance in Los Angeles the following day. The performance was highly praised by many viewers. Louis CK contacted Notaro and asked for permission to post the replay of the show. After some hesitation, she agreed to this proposal. The gig was published under the title LIVE and sold 75,000 times on the Louis CK website alone. The album was at the Grammy Awards 2014 in the category Grammy Award for Best Comedy Album nominated, subject to there but Kathy Griffin Album Calm Down Gurrl .

Notaro later underwent a bilateral mastectomy without restoring the breasts. Kristina Goolsby and Ashley York shot the documentary Tig about Notaro's illness , which premiered at the 2015 Sundance Film Festival .

Notaro has been shooting the television series One Mississippi for Amazon Video since 2016 , for which she is the lead actor, screenwriter and executive producer.
