Anna was a daughter of Landgrave Philip I of Hesse (1504–1567) from his marriage to Christina (1505–1549), daughter of Duke Georg of Saxony .

On February 24, 1544, at the age of 14, she married the three years older Palatine and Duke Wolfgang von Zweibrücken (1526–1569) and had 13 children with him. After 25 years of marriage and the premature death of her husband after only 43 years, Anna's brother Wilhelm , together with Ludwig von der Pfalz , was the guardian of their children. Wolfgang had also appointed his brother-in-law to be the executor of his will.

Around 1590 Anna donated the St. Anna Kirchhof in Heidelberg , on which a stone monument was erected in her honor 6 years later. After the St. Anna cemetery was closed in 1845, the memorial was moved to the mountain cemetery.

Anna is buried in the Protestant castle church in Meisenheim.
