Capizucchi came from a family of the Roman nobility whose members had distinguished themselves in the service of the Church. Under the aegis of Cardinal Paolo Capizucchi , his uncle, and related to Cardinals Fabio Mignanelli and Girolamo Capodiferro , he prepared for a career in the Church by studying secular and canon law. In 1535 his uncle renounced his canonical in St. Peter in his favor , even if he reserved the income. Until 1549 he was referendarius utriusque signaturae , then until 1556 Rota auditor. A decision in favor of one of his nieces earned him the goodwill of Cardinal Gian Pietro Carafa, who, after his election as Pope Paul IV , elevated him to cardinal priest on December 20, 1555 in order to increase the number of canonists in the College of Cardinals . His first titular church was San Pancrazio (January 13, 1556), then followed Santa Croce in Gerusalemme (July 6, 1562), later the titulus of San Clemente (November 7, 1565). In autumn 1556 he was appointed a member of the Inquisition Tribunal.

On July 5th, 1557 Capizucchi was appointed Bishop of Lodi , on December 21st he received episcopal ordination in Rome by Giovanni Giacomo Barba , Bishop of Terni , co- consecrators were Cesare Cybo , Archbishop of Turin , and Ferdinando Pandolfini , Bishop of Troia . Since the Spanish government under Philip II refused to give its consent (the placet ), Capizucchi was able to visit his diocese for the first time in 1563. However, he was already trying to reform his diocese from Rome. With his metropolitan, the Milanese Archbishop Carlo Borromeo , Cardinal Pius IV , he maintained good relations and a lively correspondence. Pius V granted his advisor Capizucchi the right to live in the Apostolic Palace and appointed him a member of the Cardinals' Commission, which was supposed to comment on the question of celibacy , which had not been resolved by the Council of Trent . He also sent the two reports that Capizucchi prepared in 1567 to Borromeo and asked him to make any corrections. On November 9, 1568 he succeeded the late Cardinal Vitellozzo Vitelli as prefect of the Segnatura della grazia , his death prevented him from giving the office a personal stamp.

He was buried in his titular church, San Clemente .
