Karl succeeded his father in 1755 as Prince of Anhalt-Köthen, who at the beginning of his reign suffered considerably from the consequences of the Seven Years' War . At the end of the war, Karl tried to revitalize the country economically by revitalizing agriculture and cattle breeding and by increasing the number of new settlements through tax relief. Karl distinguished himself through religious tolerance and promotion of schools. From 1765 he was a senior of the Anhalt family.

Karl took part in the War of the Bavarian Succession , where he became major general in 1779 . In 1780 he was promoted to knight of the Black Eagle Order . After participating in the campaign in Holland in 1787 , he was appointed lieutenant general in 1788 . Karl resigned from the Prussian service at the beginning of 1789 because King Friedrich Wilhelm II could not fulfill his wish for a cavalry regiment. In May 1789 he then became an Imperial and Royal Lieutenant Field Marshal and took part in the Turkish Wars as such . There the prince died of a fever near Semlin near Belgrade in 1789 and was buried in Semlin.

His son August Christian took over the government.
