Radnor Township was founded in 1682. It was part of the Welsh Tract and was named after Radnorshire in Wales . In 1717 the "Welsh Friends" established a Quaker meeting ( Radnor Friends Meetinghouse ) near the current intersection of Conestoga Road and Sproul Road in the geographic center of the community. Welsh influence declined in the late 18th century as many left the area because of high taxation. Stone monuments were erected in various locations throughout the parish in the late 20th century to commemorate the parish's Welsh heritage.

The Lancaster Pike , the first toll road in the United States linked the cities of Philadelphia and Lancaster in Pennsylvania and led by Radnor Township. It was opened in 1794. A Western Railroad has also run through the township since the early 20th century.
