Tom and his friend Luis Escobar travel to South America, where they receive the secret map to Eldorado from a priest. The drug lord Clarence Blasko, who is also trying to find the gold, pursues the two adventurers with his two female killers. When Blaskos tries to steal the card in the hotel, Tom and Luis only manage to escape with the car provided. When Blaskos attacked from a helicopter , their Land Rover was destroyed. They move on without equipment, but are caught and captured by Amazons.

When they arrive in the village of the women warriors, they discover that three more men are being held prisoner there. These are needed by the Amazons for a fertility ceremony. When two rival Amazons fight for Tom's services for the coming night on a raft , Tom saves the loser from an alligator . With that he wins the respect of the Amazon queen. In the meantime, Blasko has discovered the village and throws bombs with anesthetic gas from the helicopter. He manages to steal the card from the unconscious Tom. Before he leaves, Blasko sets the village on fire. Tom and some Amazons awaken through the smoke, they manage to save most of the still stunned female warriors. The queen confesses to knowing Blasko, Blasko was once a prisoner of the Amazons. His two companions are members of their tribe, whom Blasko made submissive with his drugs. Finally the Queen promises to show Tom the way to the seven cities. On the journey the women get closer to the men. The group successfully repels an Indian attack. Blasko has meanwhile arrived in the first golden city. A mysterious little Inca spirit threatens him and tells him to leave the treasury. When Blasko ignores the warning, the three are attacked by Indians.

When Tom comes to the temple with the Amazons, they discover that the spirit is only a reflection . The warriors can repel the following attack by the Indians. Tom discovers the captured Blasko who immediately tries to persuade him to join forces with him.
