Nikolaj Hansen was the son of the head catechist Hans Andreas Jakob Theofilus Hansen (1860–?) And his wife Emilie Bolette Juditha Motzfeldt (1865–?). His brother was Hans Hansen (1890–?). Nikolaj married on July 22, 1913 in Qeqertarsuatsiaat Ane Maria Benigne Lynge (1896–?), Daughter of the cooper Lars Christian Rasmus Vittus Lynge (1855–1928) and his wife Vitta Pouline Ane Elisabeth Margrethe Elberg (1860–1930). From the marriage, among other things, the son Hans A. Hansen (1925-1997) emerged. Nikolaj's brother was married to his wife's sister. Another sister of his wife was married to the artist Stephen Møller (1882-1909).

He and his family moved from northern Greenland to the south of Qeqertarsuatsiaat when he was young. There he lived as a hunter. In 1927 he was a member of the South Greenland State Council . The following year he was represented by Asser Berthels and then Theofilus Johnsen took the place. He died in 1935 at the age of only 43.
