The Town of Henrietta is located in southwest Wisconsin and is traversed by the Pine River , a right tributary of the Wisconsin River, which flows into the Mississippi . The intersection of the three states of Wisconsin, Iowa and Minnesota on the Mississippi is about 90 km to the west. After Illinois there are around 125 km to the south.

The Wisconsin State Highway 80 runs north-south through the town of Henrietta. In addition, the County Highways C, CC, I and II run through the area of the town. All other roads are subordinate country roads and some unpaved roads.
With Richland Airport there is a small airfield around 30 km south of the town. The nearest commercial airports are Dubuque Regional Airport in Iowa (around 150 km south-southwest), La Crosse Regional Airport (around 110 km west-northwest) and Dane County Regional Airport in Wisconsin's capital Madison (around 125 km east-southeast).
