In 1910 he became a private lecturer in Rome, and in 1911 an associate professor for Greek and Latin literature in Messina . From 1912 to 1915 he taught in the same position in Göttingen, from 1915 to 1920 he was associate professor for Greek literature at the University of Florence .

In 1924 he was promoted to full professor for Greek literature in Florence, where he was given the chair for Greek and Latin literature. His long-time colleague there was Ettore Bignone . In the same year he was elected a corresponding member of the Göttingen Academy of Sciences . In 1925 he was among the signatories of the manifesto of anti-fascist intellectuals , which Benedetto Croce had written. In 1928 he became a corresponding member of the Bavarian Academy of Sciences . From 1936 he was a member of the Accademia della Crusca , and from 1949 until his death he headed its Centro di studi di filologia italiana . The Accademia dei Lincei he was in July 1927 as a corresponding member, but was expelled in January 1946th

From 1930/1931 until 1952 he was associate professor of classical philology at the Scuola Normale Superiore in Pisa . On the occasion of the celebration of the 200th anniversary of the founding of the University of Göttingen, he was awarded an honorary doctorate.

The Department of Classical Studies at the University of Florence was named after Pasquali. He died on July 9, 1952 as a result of a traffic accident.

The Pasqualis work is of considerable importance for the redefinition of the area and of the means of philology: The fundamental work tradizione Storia della e critica del testo (1934), he theorized a new form of textual criticism that the mechanical criteria of textual criticism method Karl Lachmann recorded , but at the same time included elements from Joseph Bédier's considerations , which aimed to give greater weight to the history of the handwritten tradition and to sources as individual, historically defined objects. He was in competition with Augusto Rostagni , whom he has surpassed in effect and aftermath to this day.
