Uwe-Frithjof Haustein (born February 20, 1937 in Dresden ) is a German medic. From 1975 to 2002 he was director of the Clinic and Polyclinic for Dermatology, Venereology and Allergology at the University of Leipzig and from 1976 a full professor there. Since 1988 he has been a full member of the Saxon Academy of Sciences in Leipzig , from 2005 to 2007 he was its president and since 2005 he has been the chairman of the academy's friends' association.

Career

Uwe Frithjof Haustein is the third of four children of the couple Friedrich Paul Max Haustein (independent master furrier) and Agnes Irmgard born. Schwalm (typist / secretary). After graduating from high school at the Cross School of Dresden studied Haustein medicine at the former Karl Marx University in Leipzig (1954-1956) and then at the Medical Academy Carl Gustav Carus Dresden (1956-1959), put in 1959 his state exam , and got a doctorate. After compulsory assistantship (1960) and a general practical year (1961), specialist training took place under Heinz-Egon Kleine-Natrop in Dresden (1962–1964). In 1967 he moved to Niels Sönnichsen as a senior physician at the Jena University Hospital , where he received his habilitation in 1969 on the subject of "Physiology and pathology of local fibrinolysis in healthy and diseased skin".

From 1970 to 1975, Haustein was Deputy Clinic Director of the Charité University Dermatology Clinic , also under Niels Sönnichsen. In 1974 he was appointed lecturer. In 1975 he was appointed clinic director, in 1976 he was appointed full professor of the clinic and polyclinic for dermatology, venereology and allergology at the University of Leipzig, and in 2002 he retired. The clinic and polyclinic includes all clinical areas such as classical dermatology, venereology, allergology, occupational dermatology, pediatric dermatology, phlebology, dermatosurgery, andrology, photodermatology, dermatohistopathology and mycology. The clinic operated up to 6 wards with a maximum of 149 beds in 1975, 49 beds in 2002. In 1986 the department for experimental dermatology was founded at the clinic.

From 1984 to 1990 he was the chairman of the dermatological society of the then GDR . At the same time, he was deputy dean for research in the university's medical faculty. Haustein was an honorary member of eight foreign dermatological societies, and he was also active in the International Union against venereal Diseases and Treponematoses (IUVDT) for over 10 years. As early as 1975, an interdisciplinary consultation hour for skin cancer patients (tumor board) was launched at the University of Leipzig in order to achieve optimal care in cooperation with radiologists, surgeons, internists and other specialist clinics.

In 1988 he was elected a full member of the Saxon Academy of Sciences in Leipzig . From 1999, Haustein held management positions as deputy and later full secretary of the mathematics and natural sciences class in the Academy's executive committee, from 2005 to 2007 he was its president. In 2005, Haustein founded the Friends of the Saxon Academy of Sciences and has been its elected chairman ever since. So far, 14 prizes have been awarded to young scientists as part of research funding, as well as numerous printing and travel allowances. Since his election as a full member of the German Academy of Science and Engineering (acatech) in 2008, he has been working in its thematic network “Health Technology ”.

Since 1991, Haustein has been working as an expert in the assessment of clinical cases at the Documentation Center for Severe Skin Reactions ( Stevens-Johnson Syndrome , Toxic Epidermal Necrolysis TEN) at the Freiburg University Skin Clinic. For decades he worked as an international reviewer on issues related to work-related scleroderma .

Honors

Research areas

Haustein wrote nine monographs, including on dermatological local therapy and sexually transmitted diseases. He also wrote over 60 book chapters, over 750 scientific publications and gave over 1050 scientific lectures. In the course of his research work, Haustein supervised 18 post-doctoral qualifications as well as over 100 doctorates from doctors and scientists.

From 1990 to 2005, Haustein headed the long-term project "Human Ecological Medical Research" of the Saxon Academy of Sciences in Leipzig on questions of environment-induced systemic scleroderma, primarily through quartz dust, wound healing and tumor-matrix interactions, not least through cells -Co-cultures.

Local fibrinolysis

In addition to clinical issues, Haustein was also interested in experimental issues from the start, initially in local fibrinolysis of the skin (Habilitation 1969 in Jena). The plasminogen activator released from endothelial cells was characterized by means of the histochemical fibrin plate method (autography), biochemical investigations and animal experiments (skin transplants on mice) and influenced by means of fibrinolysis inhibitors such as para-aminomethylbenzoic acid (PAMBA).

Autoimmune diseases

At the same time, histological and electron microscopic examinations, cell physiological and immunological work followed, and later molecular biological and genetic contributions. Overall, he succeeded in developing the clinic into a center for immunodermatology, in particular with investigations into the clinical pictures of systemic and local lupus erythematosus , dermatomyositis , systemic scleroderma and bullous pemphigoid .

In the case of lupus erythematosus, the immune complex-mediated changes triggered by mouse leukemia viruses in various organ systems were first described in F1 hybrid mice as a disease model. Later, electron microscopy , particularly in, endothelium of various organs of man, tubulo-reticular structures found their viral nature could not be proved ultimately.

The immune complex cleavage by means of molar urea resulted in an increase in the titre of the autoantibodies in systemic lupus erythematosus as well as in bullous pemphigoid, which allows a better correlation between antibody titers and clinically active disease progression. The interleukin-2 receptor expression and their release to play in the pathogenesis of bullous pemphigoid a role. The reactivity of antibodies from patients on plotted epidermal extracts was compared with immunofluorescence . HLA B7 was identified as a marker for the poor response to immunosuppressive therapy. Together with complement and proteinases from neutrophils, the bullous pemphigoid antibodies mediate dermo-epidermal separation.

Progressive scleroderma

In progressive scleroderma, the acid lysosomal hydrolases were changed and there were also deviations in the content of collagen I and III, fibronectin and collagenase in the skin tissue. The collagen aminopropeptide and laminin P1 levels were found to be elevated in the serum as a sign of activation. The serum levels of chemokines, interleukins and growth factors were increased in scleroderma. The possibility of microchimerism is discussed pathogenetically. The prostacyclin analogue iloprost increases adhesion molecules and growth factors, which explain the therapeutic effect via the vascular system.

Environment-induced scleroderma

It could be shown that the environment-induced scleroderma can also be caused by chemicals, X-rays and also by quartz dust (often due to occupational reasons as a result of inadequate health and safety precautions). In the quartz dust-induced form in particular, no differences to idiopathic scleroderma with regard to clinical, serological and immunological markers were found. Here too, there was an increased expression of interstitial collagenase. Quartz is able to induce the adhesion molecule ICAM1 in endothelial cells.

Fibroblast-specific antibody

Anja Saalbach's work group at the Leipzig University Hospital found the monoclonal antibody AS02, which was found to be homologous to the Thy1 protein and showed specificity for fibroblasts . It is important for the discovery and elimination of fibroblasts. Thy1 was detected in serum from fibroblasts and endothelial cells, it also comes from polymorphonuclear leukocytes and binds to fibroblasts and endothelial cells. It exhibits adherence to neutrophils.

Tumor-matrix interaction

Fibroblasts around malignant melanoma produce increased matrix metallo- proteinase MMP as well as increased expression of adhesion molecules ICAM-1, d. H. they promote the spread and adhesion of tumor cells and thus metastasis . Thy1 ( CD90 ) and integrin avß3 ( CD51 / CD61 ) support melanoma cell adhesion to activated endothelial cells. The Mel 4B3 mRNA, which is induced in skin tumors, is regulated by growth factors (TGFß) and proinflammatory cytokines .

Permethrin in the therapy of scabies

In 1986, local therapy for scabies using permethrin (including endemics in old people's and nursing homes) and local therapy for rosacea with metronidazole were introduced. Permethrin was compared with allethrin and benzoyl peroxide , the pyrethrins and pyrethroids discussed and an algorithm for the eradication of chronic scabies crustosa and pandemics in old people's and nursing homes with permethrin and ivermectin was presented.

Personal

Hausein has been married since 1968. The couple has a daughter. Hausein has played the recorder, violin and piano since his childhood, and church organ since his youth, occasionally also in public. He often hiked in the mountains (in rope teams up to 4200 m altitude) and enjoys cycling.
