Lorenzo Quaglio learned the craft from his father Giovanni Maria Quaglio (1700–1765), who had meanwhile moved to Vienna , where Lorenzo studied at the art academy . At the age of 20, Elector Carl Theodor appointed him to the Palatinate court in Mannheim as a theater painter and architect. In 1758 the elector appointed him court architect. In 1767 Lorenzo Quaglio worked on the enlargement of the opera house in Mannheim and in 1769 he was appointed as a theater painter at the opera in Dresden . From 1772 to 1775 Lorenzo traveled to Rome , Naples and Pompeii , where he expanded his knowledge of classical art. In 1775/76 he was involved in furnishing the theater in Zweibrücken .

With the Elector Karl Theodor he came to Munich in 1777, where he initially worked for setting up operas and theater productions. In 1780 he designed the Wain Castle in what is now the Biberach district, and in 1783 the elector commissioned him to design the Lauingen town hall . After the death of Elector Carl Theodor in 1799, Lorenzo Quaglio was retired by his successor Maximilian I. His position was given to his nephew Giulio Quaglio (1764–1801).

Quaglio was related by marriage to the royal engineer-geographer Johann Adolph Sommer , the great-great-grandfather of the writer Sigi Sommer .

In the Catholic parish church of St. Afra zu Neckargerach there is a high altar designed by him around 1760, originally the main altar of the Capuchin Church in Mannheim, which was demolished in 1839 .
