In 1876, a railway company acquired land for the first time and measured it for the construction of a settlement. Shortly afterwards, at the same time as the railway line was built, the current city was built. It was named after Adrian Iselin , the mother of Adrian C. Iselin , a director of the Sioux City and St. Paul Railroad Company . A picture of the namesake still hangs in Adrian's town hall today.

John Ireland , the Archbishop of St. Paul at the time , specifically encouraged the immigration of Catholic Irish into the newly developed farmland. From his private fortune, he bought land in the vicinity of Adrian in order to make it available for the construction of farms.

The population grew from 193 in 1880 to 671 at the census in 1890 and almost doubled by 1900. At that time, however, further population development stagnated as the neighboring towns of Ellsworth, Lismore and Wilmont also grew.
