He was appointed cardinal deacon on July 1, 1517 and received the diaconate of Santi Sergio e Bacco . In 1523 he decided instead for the diaconate of Santa Maria in Via Lata . His patronage for writers and artists became particularly well known.

He served as Apostolic Administrator of Pamplona , Spain from 1520 to 1538 ; From Alessano , Italy, 1526-1531 ; From Otranto , Italy, 1526-1536; From Gerace , Italy, 1536-1538 ; briefly in 1536 from Catanzaro , Italy; 1536 to 1538 by Oppido Mamertina , Italy (abdication in favor of his biological son Ascanio Cesarini, who succeeded him from 1538 to 1542); July 6, 1537 to June 14, 1538 from Jaën ; and from 1538 until his death from Cuenca , Spain.

When the mutinous troops of Charles V sacked Rome in 1527, he was one of the cardinals who were taken hostage.

He took part in the conclaves of 1521-1522 , which Hadrian VI. chose; from 1523 , which Clement VII chose and from 1534 , which Paul III. chose.

He became Cardinal Bishop and in 1540 elected the Suburbicarian Diocese of Albano , Italy.

In 1541 he was appointed bishop of Palestrina , Italy, in whose office he died on February 13, 1542 in Rome. He was buried in his family's grave in the Church of Santa Maria in Aracoeli in Rome.
