The rowing competition in One of the Women as part of the rowing regatta of the 2016 Summer Olympics in Rio de Janeiro was held from August 6th to 13th, 2016 in the Rodrigo de Freitas lagoon . 32 athletes from 32 countries competed.

The competition, which was held over the Olympic rowing distance of 2000 meters, began with six heats with five or six teams each. The first to third placed rowers of the prelims qualified directly for the quarter-finals, while the remaining 14 boats had to go to the three hope runs . Here the first two teams qualified again for the quarter-finals and all the others for the semi-finals E / F for places 25 to 32. In the quarter-finals, four runs were carried out with six rowers each, with places 1 to 3 for semi-finals A. / B qualified, and places 4 to 6 for the semifinals C / D for 13th to 24th place. In the two rounds of the semifinals A / B the first three rowers made it to the final, while 4th to 6th place went to the B final Place 7 to 12 led. The qualification mode in the semifinals C / D and E / F ran analogously for the finals C and D or E and F for lower placements. In the final on August 13th, the best six rowers competed for Olympic medals.

The teams qualified for the next round are highlighted in light green.

The Australian Kim Brennan was the world champion of the previous year as the favorite and lived up to this role. After she had problems in the run-up due to the weather and only finished third there, she easily won her quarter-finals and semi-finals. In the final, she managed a start-to-finish victory in front of Genevra Stone from the United States and Duan Jingli from the People's Republic of China. The final was completed by Emma Twigg from New Zealand, Jeannine Gmelin from Switzerland and Magdalena Lobnig from Austria. In the B final rowed the defending champion and vice world champion Miroslava Topinková Knapková from the Czech Republic, the two-time Olympic champion Ekaterina Karsten from Belarus and the silver medalist from London Fie Udby Erichsen from Denmark.
