A year before the end of his studies, Santini signed a professional contract with the New Jersey Devils in April 2016, ending his college career. During the rest of the 2016/17 game year , the defender made his debut for the Devils in the NHL. At the beginning of the 2016/17 season , Santini found himself in the roster of the Albany Devils farm team in the American Hockey League , before he was again ordered to the New Jersey Devils at the beginning of January 2017 and was then regularly used there. He also spent the 2017/18 season alternating between the NHL and the new AHL farm team, the Binghamton Devils , before his contract was extended by three years in the summer of 2018.

In June 2019, however, Santini was handed over to the Nashville Predators along with Jeremy Davies and a second-round vote for the 2019 and 2020 NHL Entry Drafts . In return, the Devils received PK Subban .
