Dr. Victor Donovan invites the best martial artists to the ultimate martial arts tournament (DOA). The DOA winner will receive $ 10 million in prize money. The Japanese Ninja -Prinzessin Kasumi leaving her temple to join the DOA after her bodyguard Hayabusa has informed about the death of her brother Hayate, who took part in last year's DOA. Ayane, who is in love with Hayate, is tasked by the clan with killing Kasumi because she should not have been allowed to leave the clan. The US professional wrestler Tina Armstrong receives an invitation to the DOA sent on a Shuriken after she has overpowered some pirates who wanted to occupy her yacht on her boat in the South China Sea . Christie Allen gets her shuriken when she escapes police after a diamond robbery from a Hong Kong hotel. When Kasumi competes against Leon, her brother's last opponent, and wins, she realizes that Hayate cannot have died in the fight. Ayane shows up during a beach volleyball game, but is driven away by the fighters who have become friends. Meanwhile, Christie and her boyfriend Max are trying to get to the vault, which is filled with more than 100 million dollars. With the semi-finals of the tournament approaching, Hayabusa has disappeared. Donovan holds him prisoner and also brings the other semi-finalists Tina, Christie and Kasumi under his control. With the nanobots injected during the medical examination, he wants to analyze their strengths and use them for himself. He also wants to sell the program to international terrorists. In this situation, he reveals that he caught Hayate and challenges Kasumi's brother, the best fighter, to test his program to fight. He also killed Helena's father, the founder of the DOA, because he did not agree with Donovan's research.

However, Donovan's assistant and computer specialists Weatherby and Helena manage to stop the transmission of the programs to the terrorists and free the prisoners. Donovan flees the vault with the 100 million dollars and activates the self-destruction of the building complex on the island. Shortly before the whole complex self-destructs, the fighters, with the help of Hayate and Ayane, can neutralize Donovan, who dies in the explosion, and escape from the island. Kasumi returns to the palace with her new friends Tina, Christie and Helena, where they face off against the palace guards.

DOA - Dead or Alive was filmed from May 4th to July 19th in Bangkok , Guilin , Hengdian and Hong Kong . The German company Constantin Film produced the film. It premiered in September and October 2006, around ten years after the video game was released.
