The Greenville Post Office has existed since 1819, the settlement was elevated to a city in 1855, and City status was granted to Greenville in 1872.

One theory says that the city ​​of Greenville , North Carolina, named after the Revolutionary War general Nathanael Greene , was the inspiration for the name. Another theory is put forward by early settler Thomas White , who found the area "so green and beautiful". During the 1840s a number of citizens of Bond County on helping Underground Railroad escaped slaves from the South to freedom. The slaves often came to Bond County from Missouri . Rev. John Leeper hid his work for the Underground Railroad under the guise of the mill he operated. Doctor Henry Perrine , a doctor near Greenville , also helped with the underground railroad's hidden activities.

In 1855, Greenville College was founded under the then name Almira College .

In 1858, both Abraham Lincoln and Stephen Douglas made speeches in Greenville in the congressional election .

On November 21, 1915, the Liberty Bell came through Greenville on its way back across the country from the World's Fair in San Francisco to Pennsylvania . After this trip, the bell remained in Philadelphia and was never moved from there again.

During the Great Depression 500 demonstrators marched on April 18, 1934 for "Illinois Emergency Relief Commision" ( Commission for Notfürsorge of Illinois ), to protest the slow pace of distribution of relief supplies to the state and the federal government.

Ronald Reagan came to Greenville during the presidential election campaign in 1980 and gave a speech on the street in front of the courthouse.
