As the youngest son of Camillo, the third Duke of Sermoneta from the noble family of the Caetani di Sermoneta , and his second wife Flaminia Savelli, Niccolò was a cousin of Alessandro Farnese, Pope Paul III. On March 13, 1538, he was published as a cardinal in the consistory and on April 16 of the same year he received the cardinal's hat and the title diaconia of San Nicola in Carcere . From March 9, 1552 he was cardinal priest pro illa vice with Sant'Eustachio as the titular church .
In January 1577 he became treasurer of the Holy College of Cardinals , a position which he held until the beginning of the following year.
He died in Rome on May 1, 1585 at the age of 59 and was buried in the basilica of the Holy House in Loreto in the atrium of the Sacred Heart Chapel. The design for the tomb, which was commissioned by the cardinal himself during his lifetime and which was completed in 1580, comes from Francesco Capriani , also Francesco da Volterra, the marble figures come from Fides (left) and Caritas (right), like the design for the bronze statue of the Cardinal by Giambattista della Porta , the bronze casting was carried out by Antonio Calcagni and Tiburzio Vergelli .
