The Kakadu Highway begins in Pine Creek, a village 91 km north of Katherine and 230 km south of Darwin , where it branches off in a northeastern direction from the Stuart Highway.

After 48 km the road reaches the Mary River Roadhouse , which is located on the river of the same name and at the same time forms the entrance to the Kakadu National Park.

On its way the highway offers access to many tourist highlights of the Kakadu National Park. Smaller roads branching off the Kakadu Highway lead to the following locations: Gunlom Falls , Maguk Swimming Pools , Gagudju Lodge and the Warradjan Aboriginal Cultural Center , Jim Jim Falls , Twin Falls and Nourlangie Rock. There are also numerous lesser-known attractions along the Kakadu Highway. There are several campsites and lookout points along the highway. The Jim Jim Road , which branches off near Jim Jim Billabong , leads towards Darwin.

Eventually the Kakadu Highway reaches its terminus in Jabiru. This is the main town in Kakadu National Park. In addition to the Kakadu Highway, the Arnhem Highway (S36) meets here from the west , the Strait of Ubirr and the Border Store from the north and the Strait of Jabiru East, the airport and the Ranger uranium mine from the east .

The highest point on the highway is at 259 m, the lowest at 8 m.
