In 2016 she reached the final of an ITF tournament in the $ 60,000 category in Essen and then qualified for a Grand Slam tournament for the first time at the US Open , but failed in the first round.

The following year, Muchová stood in front of a home crowd in Prague, first in the final of a tournament in the $ 80,000 category, which she lost to her compatriot Markéta Vondroušová , before qualifying for the main draw of a WTA tournament in Seoul for the first time ; in the first round she lost to Priscilla Hon in three sets. In 2018, Muchová reached three more finals on the ITF tour, first at the $ 25,000 tournament in Altenkirchen , then at the $ 60,000 tournament in Croissy-Beaubourg and finally at her home tournament in Olomouc in the $ 80,000 category, which she lost all. At the US Open , she made international attention for the first time when she went straight to the third round in her first successful qualification for the main draw of a Grand Slam tournament after victories against Dajana Jastremska and Garbiñe Muguruza she first had to admit defeat to Ashleigh Barty .

In 2019 Muchová had an international breakthrough. After qualifying in Doha at the beginning of the season, she advanced to the quarter-finals and thus reached the top 100 in the tennis world rankings . In Prague she was in her first final on the WTA Tour, which she lost to Jil Teichmann . Her greatest career success up to then, however, she achieved at Wimbledon , where she moved into the quarter-finals after a 13:11 win in extra time in the third set against her compatriot Karolína Plíšková , her first success against a top 10 player; in the round of the last eight she lost to Elina Switolina . Following another third round participation at the US Open , in which she had to admit defeat to Serena Williams , Muchová was able to celebrate her first WTA title in Seoul after a final victory over Magda Linette . By reaching the semi-finals at the Kremlin Cup in Moscow, in which she was defeated by Anastassija Pavlyuchenkova , she qualified for the first time for the WTA Elite Trophy 2019 in Zhuhai. There she came after two wins in the group stage against Alison Riske and Sofia Kenin in the semifinals, in which she was defeated by the eventual winner Aryna Sabalenka . Nevertheless, Muchová finished the season in 21st place and thus achieved her highest world ranking position so far.

In 2019 she made her debut in a 4-0 playoff win against Canada in singles for the Czech Fed Cup team , where she won her game against Rebecca Marino .
