First Laxalt played in a club in the Barrio and then at Cerro Junior. He joined Defensor Sporting at the age of nine and went through the youth teams. The fast midfielder began his professional career with Defensor, where he made his debut in the Uruguayan Primera División on September 1, 2012 against the Montevideo Wanderers . On February 24, 2012, he scored his first league goal in a 1-0 win over Nacional Montevideo .

For the 2013/14 season Laxalt moved to the Italian Serie A at Inter Milan . He signed a five-year contract until June 30, 2018. In the same transfer period, however, he was loaned to Inter's club FC Bologna in August 2013 as part of the transfer from Saphir Taïder to Inter until the end of the season. The Bolognese also secured an option for 50 percent of the transfer rights. There he made his debut on September 25, 2013 in the Serie A game against AC Milan . In his first appearance, he contributed two hits to the 3-3 draw. He played a total of 15 league games (two goals) for Bologna in the 2013/14 season.

For the 2014/15 season , Laxalt was loaned to FC Empoli . The receiving club received a purchase option and Inter received a return option. In the first half of the season he made four first division appearances at Empoli (no goal).

At the end of January 2015, after Inter terminated the loan agreement with Empoli, he was loaned out to CFC Genoa , trained by Gian Piero Gasperini , for 18 months . By the end of the 2015/16 season , which CFC Genoa finished as sixth in the table, he was used there eight times (no goal) in Serie A. For the 2016/17 season , CFC Genoa acquired the transfer rights to Laxalt. During the season he played a total of 36 first division games and scored one goal.

On August 17, 2018, Diego Laxalt moved to AC Milan . He signed a four-year contract until June 30, 2022. In his first year, he played 20 league games.

At the end of August 2019, he was loaned to the league rivals FC Turin for the 2019/20 season with a subsequent purchase option. This loan was canceled early in late January 2020, shortly before the end of the transfer window, and Laxalt returned to the AC Milan squad.
