The Town of Burke is located in southern Wisconsin, in the northern and northeastern suburbs of its capital, Madison . The intersection of the three states of Wisconsin, Iowa and Minnesota, located on the Mississippi , is about 195 km to the west-northwest; to Illinois it is around 80 km south.

The Interstate Highways 39 and 90 , which run on the same route, form the eastern bypass of the city of Madison and lead in part through the area of the Town of Burke. The US Highway 151 , the north-eastern arterial road of Madison, runs in part by the area of the Town of Burke. The US Highway 51 runs in a north-south direction by the area of the town. All other roads are subordinate country roads and some unpaved roads.
For freight transport two railway lines to run Canadian Pacific Railway belonging SOO Line Railroad through the area of the Town.
The nearest airport is Dane County Regional Airport in Wisconsin's capital Madison (just across the town's border).
